AOH1996 is an experimental anticancer drug, a small molecule inhibitor of proliferating cell nuclear antigen (PCNA), that entered phase I clinical trials for the treatment of breast cancer in August 2023. Because it kills cancer cells in many types of cancer, AOH1996 hopes the drug could one day be used to treat cancers of the breast, prostate, brain, ovary, uterus, skin and lung. "No one approached PCNA as a cure because it was considered 'unavoidable,'" said Long Gu, a City of Hope researcher and director of the study, published in Cell Chemical Biology.

Another benefit of AOH1996 is its ability to make cancer cells more susceptible to drugs that damage DNA or chromosomes, such as the antibiotic cisplatin. This means that the new drug could one day be an important tool in the development of new combinations and chemotherapy.

The drug AOH1996 was named after a daughter of cancer patient Anna Olivia Healey, who died in 2005. The Pill's story began when Linda Malkas started with Anna's family in 2005, after her daughter had a younger battle with neuroblastoma, a deadly form of cancer. According to the American Cancer Society, neuroblastoma starts early in the brain, usually in the embryo or foetus. When Linda met Anna's father at the end of her life. She asked her father if there was anything he could do about neuroblastoma.

Anna's father immediately wrote down Linda's lab for $25,000. That moment changed Linda's life forever. AOH1996 represents the culmination of two decades of research by Linda Malkas and City of Hope. The new drug works by targeting a protein called PCNA (proliferative cell nuclear antigen). Modified versions of PCNA help cancer cells multiply and grow. "The fact that PCNA is specifically mutated in cancer cells allows us to develop drugs that target only PCNA in cancer cells," said Linda Malkas, as the healthy body leaves the normal brain. She also said that cancer drugs are like a storm that shuts down major airports and all flights, but "only on airplanes with cancer." Drugs AOH1996 is currently in phase 1 clinical trials at City of Hope. In previous experiments, AOH1996 selectively kills cancer cells by interrupting the cell reproduction cycle. The A.N.N.A. Fund (Anna Needs Neuroblastoma Answers) is a charity that has raised over $400,000 for neuroblastoma research since 2002. Anna's father didn’t know of how to appreciate the love and support shown to our daughter by her friends, family and strangers. Before Anna was born, Anna's father, Steve, wrote on the family's website that Anna worked with her 56-year-old mother, Barb Healey.

“Thanks to the awareness and financial support of people like Linda, we will be able to pass on the same love and care to other families who are in the same situation as us,” she said. “We couldn’t save Anna but we could save many others. When you see me, I always say, I have a 9-year-old girl sitting on my right shoulder. She's my touchstone," she added.
